The basement membrane is the key adhesion zone between the basal layer of epidermal keratinocytes and the dermis. Its integrity is of great significance for maintaining the youthfulness of the skin. Type IV collagen is synthesized in the rough endoplasmic reticulum and secreted into the basement membrane through the Golgi apparatus. It plays a pivotal role in the basement membrane. Type IV collagen is tightly bound to a variety of components to build a stable basement membrane network. The triple helical region of type IV collagen forms a tight connection with the G2 domain of nidogenin; at the same time, nidogenin binds to perlecan and laminin. Fibroblasts are one of the main cells in the dermis. Skin aging phenomena such as decreased amounts of elastin and collagen, and increased wrinkles are believed to be related to decreased numbers and decreased vitality of fibroblasts. The dermis is divided into the papillary layer and the reticular layer. The papillary layer fibroblast subpopulation is the papillary fibroblast, and the reticular layer fibroblast subpopulation is the reticular fibroblast. The two types of fibroblasts differ in cell morphology, function, and secreted substances, which affect the interaction between fibroblasts and surrounding cells, especially keratinocytes. Skin aging involves a variety of changes, among which papillary fibroblasts are gradually replaced by reticular fibroblasts, which will affect the morphological structure and function of the basement membrane, thereby accelerating skin aging.
